- What's the difference between using "when to execute" on a destination or on the layout.
The only difference is performance. If you use it on the layout, Optio will create the layout first and then looks at the when to execute. When you use the when to execute on the destination, it will act on the whole part.

- Is there some size limit for Functions/Code before/Code After? The Code dialog does not open anymore. Copying data does not work properly.
Welcome to the wonderful world of the Win32 programmer. It has to do with internal settings Microsoft has changed in order to provide you with a new version of theire OS. When you have trouble with the size of the code, The sizelimit should be somewhere around 32Kb (for Windows XP).
